ABSTRACT

Diabetic retinopathy (DR) is a common microvascular complication of diabetes mellitus, with capillary occlusion, microaneurysms, and neovascularization as main pathological features, and its pathogenesis has not been fully understood.The existing clinical treatment can not cure DR, therefore, the study of the pathogenesis of DR is still a hot topic and difficult problem.Current studies have suggested that oxidative stress response is one of the important mechanisms for the development of DR, which promotes the production of excessive reactive oxygen species to induce retinal cell dysfunction and apoptosis.Imbalance of levels of nitric oxide (NO) from different types of sources in the retina under hyperglycemic stimulation also plays an important role in pathological changes, such as capillary damage, blood-retinal barrier disruption and neovascularization in DR.At the same time, NO interacts with oxides produced by oxidative stress to promote further retinal damage in DR, leading to retinal morphology abnormality and dysfunction, which ultimately cause irreversible blindness in patients.Research status of NO and oxidative stress in the pathogenesis of DR from pathophysiological changes of DR, DR and oxidative stress, DR and NO, and the role of NO in oxidative stress were reviewed in this article.

ABSTRACT

Objective To investigate the effect of advanced oxidation protein products (AOPPs) on human renal tubular epithelial cells(HK-2) autophagy.Methods HK-2 cells were stimulated with AOPPs.RT-qPCR and Western blot were used to determine the expression of autophagy related protein LC3-Ⅱ/LC3-Ⅰ,Beclin1 and p62;Western blot was utilized to examine the activation of p38 MAPK pathway.Then p38 MAPK inhibitor (SB203580) was added and co-processed with AOPPs.The change of autophagy was observed Also,autophagy inducer rapamycin was added and co-processed with AOPPs.RT-qPCR and Western blot were used to detect the expression of cell cycle inhibitory protein p27.The cell total protein level was detected by the bicinchoninic acid (BCA) method.The hypertrophy change was observed.Results AOPPs down-regulated the expression of LC3-Ⅱ/LC3-Ⅰ and Beclin1,up-regulated expression of p62 and activated p38 MAPK pathway;in comparison with the AOPPs alone treatment group,the expression of LC3-Ⅱ/LC3-Ⅰ and Beclin in the SB203580 co-processing group was increased,while p62 was decreased;the p27 expression and cells total protein in the sirolimus co-processing group were down-regulated.Conclusion AOPPs inhibits the autophagy of HK-2 cells by activating p38 MAPK pathway and the autophagy inhibition participates in HK-2 cell hypertrophy.

ABSTRACT

Diabetic retinopathy (DR) is the most serious and common blinding ocular complication in all microangiopathy of diabetes mellitus.The occurrence of DR associated with a variety of factom,the pathogenesis has not yet been completely clear,and there are several theories.In recent years,the relationship between the occurrence of DR and sex hormone levels become a hot ophthalmic research,research shows that the prevalence of DR significantly increased in postmenopausal women,in which estrogen change plays the main role.Learning connection between estrogen and DR will contribute to the clinical diagnosis and treatment of DR.This article reviews recent research advances in the relationship between estrogen and DR.

ABSTRACT

Objective:To analyze the clinical characteristics of interstitial pulmonary fibrosis (IPF) in patients with Rheumatoid arthritis (RA). Methods: We retrospectively analyzed 198 RA patients with or without IPF. Characteristics of RA-IPF in clinical and lab data were analyzed. Age, duration of disease,clinical and laboratory parameters, history of smoking and medicine were compared between the patients with and without IPF. Results: (1) Among the 198 RA patients, 15.2% (30/198) were found with IPF. 100% RA-IPF patients had HRCT findings. However, 63.3% (19/30) had positive findings in chest X-ray, and only 46. 7% ( 14/30) had the complaints of cough and short breath. Velcro rales were found in 50.0% (15/30) patients with IPF and no acropachy occurred. Only one patient suffered from hypoxemia. IPF presented after the joint symptoms in most patients. (2 ) RA-IPF patients were older than those without IPF [(65.50±9.71) vs (55.22±12.98) years, P<0.01]; Higher positivity of anti-keratin antibodies ( AKA) were found in RA-IPF compared to patients without IPF (61. 5% vs 35.9% , P =0.014). Furthermore, the levels of anti-cyclic citrullinated peptide (CCP) antibody were significantly higher in RA-IPF [(4.38±2.08) vs (3.20±2. 12) , P =0.01]. No differentiation of duration of disease, history of smoking and medicine, IgM rheumatoid factor, IgG rheumatoid factor, anti-nuclear antibody, anti-SSA antibody and levels of immunoglobins and complements were found between the two groups of RA patients with and without IPF. Conclusion: The clinical symptoms of IPF in RA patients are mild and more common in older patients. AKA and anti-CCP antibody might be important antibodies associated with RA-IPF.
